These diagnostic discs are constructed from high-quality, absorbent filter paper precut into small circles. They come pre-treated with an analytical redox indicator reagent—most commonly Kovacs reagent (N,N,N',N'-tetramethyl-1,4-phenylenediamine) or N,N-dimethyl-p-phenylenediamine oxalate combined with α-naphthol. To perform the test, a clinician uses a plastic loop or sterile wooden stick to pick up an isolated bacterial colony from an agar plate and rubs it directly onto the disc.
